      目的:探究韩式治疗仪干预大鼠孤独症仔鼠的作用分子机制。方法:通过大鼠合笼见阴栓为妊娠第1d(E1),孕鼠在 E12.5d 时(孕中期),模型组孕鼠腹腔注射丙戊酸钠 600 mg/kg,出生后3周开始进行韩式治疗仪干预“长强穴”和“太溪穴”韩式治疗仪治疗7天,在干预前进行模型组与正常组的行为学观察,干预结束后,处死各组大鼠,取脑组织,进行 HE染色,qPCR 检测其中IL-1β、TNF-a、IL-4、p53 mRNA的表达情况。结果:行为学分析显示:模型组子代鼠发育明显落后:体重偏低、睁眼时间延长,1个月内病死率和畸形率高于对照组,修饰行为、群集性活动少于对照组,攻击性行为增加( P <0.05)。与正常对照相比较,模型组理毛时间少、次数少于对照组,差异有统计学意义(P <0.05) ; 模型组出现撕咬、拳击、后顶、脚蹬、攀压等搏斗动作,攻击行为间断进行,对照组未出现攻击行为;模型组群集活动正常对照持续时间短、次数少于对照组,差异有统计学意义( P <0.05) 。HE染色结果显示,正常对照组海马细胞结构完整正常, 在浦肯野细胞层中,主要由单层大梨形细胞组成,胞浆嗜酸性,胞核呈泡状,浦肯野细胞结构正常。大鼠孤独症模型组海马细胞减少,浦肯野细胞层细胞减少,部分炎症细胞浸润。模型+韩式治疗仪干预组海马细胞结构清晰正常,浦肯野细胞层细胞部分恢复增生,凋亡细胞减少,正常细胞增多。qPCR检测模型组大鼠脑组织IL-1β、TNF-a、IL-4、p53 mRNA表达上升,韩式治疗仪干预后IL-1β、TNF-a、IL-4、p53 mRNA表达下降。结论 韩式治疗仪干预可能通过抑制炎症反应,对大鼠孤独症模型仔鼠具有一定的治疗作用。

      Objective: To explore the molecular mechanism of HANS therapeutic instrument in the intervention of autistic spectrun disorder (ASD) rat offspring. Methods: The rats in the model group were injected with 600 mg/kg sodium valproate peritoneally at E12.5d (second trimester),and were treated with HANS therapy instrument at Changqiang Point and Taixi Point for 7 days from 3 weeks after birth. Behavioral observation was conducted between the model group and the normal group before intervention. After the intervention, the rats in each group were killed, and the brain tissues were taken for HE staining,and the mRNA expressions of IL-1β、TNF-a、IL-4 and p53 were detected by qPCR. Results: The behavioral results showed that compared with the normal pair,the offspring rats in the model group had a significantly lower body weight, a longer time to open eyes, a higher mortality rate and malformation rate within 1 month, less modification behavior and swarming activity, and an increased aggressive behavior (P < 0.05), the grooming time and frequency of the model group were less than that of the control group, and the difference was statistically significant (P < 0.05).In the model group,there were biting, punching, back top, foot pushing, pressing and other fighting actions,and the aggressive behavior was intermittent for 3-5 min, while no aggressive behavior was observed in the control group. The duration and frequency of cluster activity in model group was shorter than that in control group, and the difference was statistically significant (P < 0.05). HE staining showed that the structure of hippocampal cells in the normal control group was complete and normal,and the Purkinje cell layer was mainly composed of single large pyriform cells with eosinophilic cytoplasm and bulbous nucleus, and Purkinje cell structure was normal. In the rat model group of autism,hippocampal cells decreased, Purkinje cell layer cells decreased,and some inflammatory cells infiltrated. In the treatment group of model HANS therapy instrument, the hippocampal cell structure was clear and normal,some cells in the Purkinje cell layer recovered proliferation, apoptotic cells decreased, and normal cells increased. The mRNA expressions of IL-1β、TNF-a、IL-4 and p53 in the brain tissue of the model group were increased by Qpcr,while the mRNA expressions of IL-1β、TNF-a、IL-4 and p53 were decreased after the intervention of the HANS treatment instrument. Conclusion: The intervention of HANS therapy apparatus may have a certain therapeutic effect on the autism model of rat offspring by inhibiting the inflammatory response.
