      目的 亲环素A(cyclophilin A,CyPA)在糖尿病大鼠动脉粥样硬化进展中的作用。 方法 实验设置对照组、动脉粥样硬化组(AS组)、糖尿病动脉硬化组(DM组)。每组随机分配雄性Wistar大鼠10只。对照组只给予基础饲料。AS组高脂饮食+腹腔注射维生素D3。DM组腹腔注射维生素D3,高脂饲养4周后行糖尿病造模并继续高脂饲养。16周后检测三组大鼠血糖、动脉硬化指标及炎性因子,观察大鼠腹主动脉病变,检测各组大鼠动脉CyPA、p-ERK1/2及NF-kB的表达。 结果 与AS组相比,DM组大鼠多消瘦,体重下降,血糖升高,血清炎性因子增加,动脉硬化指数ASI显著增高。AS组及DM组大鼠腹主动脉粥样病变明显,且血管内皮损伤,炎性细胞浸润,粥样斑块,钙化斑逐渐加重。与AS组相比,DM组大鼠血管壁CyPA、p-ERK1/2和NF-kB蛋白表达显著增加。结论 CyPA蛋白在糖尿病大鼠病变血管壁中显著表达,可能参与糖尿病动脉血管粥样硬化形成和发展。

      Objective To investigate influences of cyclophilin A (CyPA) on atherosclerosis in diabetic mice. Methods A total of thirty SPF male wistar mice were randomly divided into three groups (i.e., control group, atherosclerosis group (AS group) and diabetic group (DM group)), with 10 mice in each group. The control group were fed with base diet, and given intraperitoneal injection of equal normal saline (NS). The AS group were fed with high cholesterol diet, and given intraperitoneal injection of a single dose of Vitamin D3 600,000 IU per kg body weight. A total of ten DM group male wistar mice were given intraperitoneal injection of a single dose of Vitamin D3 600,000IU per kg body weight, and were fed with high cholesterol diet for four weeks, and were rendered diabetic by intraperitoneal injection of streptozotocin at a dose of 50mg/kg. After 16 weeks, ELISA was used to detect blood glucose, arteriosclerosis indicators, and inflammatory factor levels in the serum of mice. At the same time, HE staining was used to observe the morphological changes of abdominal aortic lesions in mice, and immunohistochemistry and Western blot techniques were used to detect the expression of CyPA, phosphorylated extracellular signal regulated kinase1/2 (p-ERK1/2), and nuclear factor-k-gene binding (NF-kB) in the arteries of mice in each group. Results Compared with the AS group, the DM group mice tended to lose more weight, and more significantly decrease in body weight, while increase blood sugar levels, serum inflammatory factors, and arteriosclerosis index (ASI) (all P<0.05, DM group versus AS group). The atherosclerotic lesions of abdominal aorta in AS group and DM group were obvious, and the vascular endothelial injury, inflammatory cell infiltration, atherosclerotic plaque, and calcified plaque were gradually aggravated. Compared with the AS group, the expression levels of CyPA, p-ERK1/2, and NF-kB proteins in the vascular wall of DM group mice were more significant, and the difference was statistically significant (P<0.05, DM group versus AS group). Conclusions CyPA proteins are significantly expressed in the diseased vascular wall of diabetes rats and participate in the formation and development of atherosclerosis, which may be one of the ways of the progression of diabetes vascular disease.
