      目的:观察头针对局灶性脑缺血再灌注后大鼠脑组织内环氧化酶-2(Cyclooxygenase-2, COX-2)表达的影响,探讨头针对脑缺血再灌注后损伤的保护机制。方法:将100只SD大鼠随机分为正常对照组、假手术组、模型组和头针组,根据缺血再灌注时间点,再将模型组和头针组各分为缺血再灌注后6h、24h、48h、和96h组,每组各10只。采用线栓法构建大脑中动脉闭塞脑缺血模型,头针作用于顶颞后斜线和顶颞前斜线30分钟,分别采用HE染色观察各组大鼠脑组织内炎性浸润情况,采用荧光定量PCR、酶联免疫吸附(ELISA)法检测各组大鼠脑缺血区COX-2mRNA和COX-2蛋白表达的变化。结果:(1)与模型组相比,头针组大鼠缺血再灌注后24、48、96h神经功能缺损评分(National Institute of Health sroke scale, NIHSS)明显降低,差异均有统计学意义(P < 0.05)。(2)与模型组相比,头针组大鼠脑缺血组织炎性细胞表达减少。(3)头针组大鼠脑缺血组织COX-2 mRNA表达明显减少,与模型组相比,缺血再灌注6h、24h、48h、96h后差异具有统计学意义(均P < 0.05)。(4)头针组大鼠脑缺血组织COX-2蛋白表达明显减少,与模型组相比,缺血再灌注24h、48h、96h后差异具有统计学意义(均P < 0.05)。结论:头针治疗能通过抑制脑缺血区COX-2 mRNA及其蛋白表达,促进脑缺血再灌注后损伤的康复。

      Objective: To observe the effects of scalp acupuncture on the expression of cyclooxygenase-2 (COX-2) in brain tissue of rats after cerebral ischemia-reperfusion injury, so as to investigate the protective mechanisms of scalp acupuncture on improving injury after cerebral ischemia- reperfusion. Methods: One hundred SD rats were randomly divided into the normal control group, the sham operation group the model group and the scalp acupuncture group, then rats in the model group and the scalp acupuncture group were further divided into 6h, 24h, 48h and 96h groups after ischemia reperfusion, with 10 rats in each group. The middle cerebral artery occlusion (MCAO) rat model was established by the improved thunking method. In the scalp acupuncture group, acupuncture needles were inserted into apiotemporal posterior oblique line and apiotemporal anterior oblique line for 30 minutes. We observed the changes in inflammatory infiltration of ischemic tissue by using HE staining and detected the expression changes of COX-2 mRNA and protein in ischemic tissue after treated with scalp acupuncture by using PCR and ELISA. Results: (1) Compared with the model group, NIHSS scores in the scalp acupuncture group were significantly decreased at 24h, 48h and 96h after ischemia reperfusion (all P < 0.05). (2) (3) The expression of COX-2 mRNA decreased in ischemic tissue after treated with scalp acupuncture, compared with the model group, significantly difference were observed at 6h、24h、48h and 96h after ischemia reperfusion (all P<0.05). (4) The expression of COX-2 protein decreased in ischemic tissue after treated with scalp acupuncture, compared with the model groups, significantly difference were observed at 24h、48h and 96h after ischemia reperfusion (all P<0.05). Conclusion: The scalp acupuncture could promote the recovery of cerebral ischemia-reperfusion injury by inhibiting the expression of COX-2 mRNA and protein in the ischemic area.
