阿尔茨海默病(Alzheimer’s disease,AD)是一种最常见的神经退行性疾病,主要临床表现为损伤性
的、不可逆转的认知功能障碍。睡眠-觉醒节律紊乱是AD患者的常见症状,然而大量研究表明早在AD行
为症状发生之前睡眠-觉醒节律紊乱已经存在。正常的睡眠-觉醒节律有利于β淀粉样蛋白(Amyloid-β-protein,Aβ)和tau蛋白的代谢。然而,当睡眠-觉醒周期被打乱,中枢神经系统对有害代谢物的清除就会减
少。此外,睡眠-觉醒节律紊乱也可导致神经炎症和中枢神经系统氧化应激的产生,加速认知的损害。因
此,本综述主要探讨睡眠-觉醒节律紊乱与AD发生发展的关系。 |

Alzheimer’s disease (AD) is the most common neurodegenerative disease, primarily characterized
by detrimental and irreversible cognitive dysfunction. Sleep-wake rhythm disturban is a common symptom in
AD patients, and extensive researches have shown that sleep-wake rhythm disturban predates the onset of
behavioral symptoms of AD. The normal sleep-wake rhythm facilitates the metabolism of Amyloid-β-protein (A
β) and tau protein. However, when the sleep-wake cycle is disturbed, the central nervous system’s ability to
clear harmful metabolic waste diminishes. Moreover, sleep-wake rhythm disturban can also lead to
neuroinflammation and oxidative stress in the central nervous system, accelerating cognitive impairment.
Therefore, this review primarily explores the relationship between the disturban of the sleep-wake cycle and the
development of AD. |
