      神经退行性疾病是近年引发患者脑组织功能障碍的主要原因和第二大死亡原因,对全球人类健康造成了严重威胁。血脑屏障(Blood-Brain Barrier, BBB)具有保护中枢神经系统维持大脑稳态的功能。然而,神经炎症会导致BBB功能障碍,进而诱导神经胶质细胞过度活跃,进一步破坏BBB功能的正常运作,最终引发神经退行性疾病。在众多疗法中,干细胞疗法可以精准高效地治疗神经退行性疾病。尽管干细胞治疗神经退行性疾病前景广阔,但仍面临许多挑战,例如干细胞的安全性和有效性、移植后的存活和整合、免疫排斥反应等。在这篇综述中,我们系统探讨了干细胞疗法通过降低炎症反应治疗神经退行性疾病的可能性和发展前景。

      Neurodegenerative diseases have emerged as a leading cause of brain dysfunction and the second major cause of mortality in recent years, posing a significant threat to global health. The blood-brain barrier (BBB) plays a pivotal role in maintaining central nervous system (CNS) homeostasis by protecting the brain from harmful substances. However, neuroinflammation can trigger BBB dysfunction, leading to hyperactivation of glial cells, which further exacerbates BBB impairment and ultimately contributes to neurodegeneration. Among various therapeutic strategies, stem cell-based therapy has shown remarkable precision and efficacy in treating neurodegenerative disorders. Despite its promising potential, several challenges remain, including ensuring the safety and efficacy of stem cells, promoting their post-transplantation survival and integration, and overcoming immune rejection. In this review, we systematically examine the therapeutic potential and future prospects of stem cell therapy in mitigating neurodegenerative diseases through anti-inflammatory mechanisms.
