阿尔茨海默病(Alzheimer's disease,AD)是一种常见的神经退行性疾病,主要表现为严重的记忆丧失
和认知功能衰退。然而,其发病机制复杂且尚未完全阐明。近年来的研究表明,铁死亡(ferroptosis)作为一
种程序性细胞死亡方式,在AD的发病和病程中发挥了关键作用。铁死亡主要由细胞内铁超负荷引发的脂
质过氧化所驱动,因此,AD中的脂质代谢紊乱显著影响细胞对铁死亡的敏感性。本文综述了脂质代谢紊
乱在AD中如何调节铁死亡,包括脂质合成异常、脂质过氧化程度增加、脂质储存与降解改变,以及脂质β氧
化减少等对铁死亡敏感性的具体影响。此外,文章还探讨了常见抗氧化剂在调节铁死亡中的潜在治疗效

Alzheimer's disease (AD) is a common neurodegenerative disorder characterized by severe memory
loss and cognitive decline. However, its pathogenesis is complex and not fully understood. Recent research has
highlighted ferroptosis, a form of programmed cell death driven primarily by intracellular iron overload and
subsequent lipid peroxidation, as playing a critical role in the onset and progression of AD. Dysregulation of
lipid metabolism in AD significantly influences cellular sensitivity to ferroptosis. This review discusses how
lipid metabolism dysregulation modulates ferroptosis in Alzheimer's disease, including the effects of abnormal
lipid synthesis, increased lipid peroxidation, altered lipid storage and degradation, and reduced lipid β-oxidation
on ferroptosis sensitivity. Additionally, the review explores the potential therapeutic effects of common
antioxidants in regulating ferroptosis. These discussions provide a crucial theoretical foundation for a deeper
understanding of AD pathogenesis and the development of novel therapeutic strategies. |
