      目的:通过孟德尔随机化研究探索阿尔兹海默病领域的热点话题及发展趋势,为治疗阿尔兹海默病提供新方向与新思路。 方法:检索Web of Science核心合集数据库应用孟德尔随机化研究阿尔兹海默病的相关英文文献,检索时间为建库至2024年3月,并用CiteSpace 6.2.R4对发表文献的作者、国家、机构、文献共被引、关键词共现、关键词聚类情况进行可视化分析并绘制知识图谱。利用智慧芽全球新药情报数据库查询与阿尔兹海默病相关的生物标志物的药物研发情况。 结果:去重后共纳入347篇英文文献,年度发文量呈现上升趋势。英国学者Ben-shlomo,Yoav 中介中心性最高;发文量前3位的国家是中国、美国和英国,中介中心性排名前3位的国家依次是波兰、法国和加拿大,中国发文量最高,但仅与美国和荷兰存在合作;发文量前3位的机构是伦敦大学、哈佛大学和复旦大学;关键词聚类图中,“#0 risk factors(危险因素)”与AD的生物标志物关系最紧密,其中APP和TAU是更具潜力的药物靶点。 结论:孟德尔随机化研究阿尔兹海默病主要在中国、英国和美国开展,中国不仅是发文量做高的国家,Yu, Jin-Tai亦是发文量最高的学者。主要热点是研究AD发病的危险因素,如脑脊液中淀粉样蛋白等,且相关研究发现APP和TAU是更为重要的生物标志物及药物靶点。

      Objective: To explore the hot topics and development trends in the field of Alzheimer's disease through Mendelian randomization research, and to provide new direction and new ideas for the treatment of Alzheimer's disease. Methods: The Web of Science Core Collection was searched for English literature on the study of Alzheimer's disease using Mendelian randomization,the search time is from the establishment of the database to March 2024,and CiteSpace 6.2.R4 was used to visually analyze the authors, countries, institutions, co-citation, keyword co-occurrence and keyword clustering of published literature, and draw a knowledge graph.Use PharmSnap Global New Drug Database to query the drug development status of biomarkers related to Alzheimer's disease. Results: A total of 347 English documents were included after removing the duplicates, and the annual number of articles showed an upward trend., British scholar Ben-shlomo, Yoav has the highest intermediary centrality; the top 3 countries of number of publications are China, the United States and the United Kingdom;the top 3 countries of intermediary centrality are Poland, France and Canada.China has the highest number of publications, but only cooperation with the United States and the Netherlands; The top 3 institutions in terms of the number of papers published are the University of London, Harvard University, and Fudan University.In the keywords clustering graph, “#0 risk factors” is the most closely related to the biomarker of AD, among which APP and TAU are the more potential drug targets. Conclusion: Mendelian randomization study of Alzheimer's disease is mainly carried out in China, the United Kingdom and the United States. China is not only the country with a high number of publications, but also Yu and Jin-Tai are also the scholars with the highest number of publications. The main focus is to study the risk factors of AD, such as amyloid in cerebrospinal fluid, and related studies have found that APP and TAU are more important biomarkers and drug targets.
