目的:观察失神经支配后大鼠面肌形态和肌细胞收缩相关蛋白的变化。方法:对健康成年SD大鼠23只进行研究,左侧切断面神经出茎乳孔处主干(断伤侧),右侧作相同切口仅暴露面神经(对照侧)。2月后取两侧鼻唇提肌,进行Masson染色观察两侧肌纤维形态学变化,包括肌细胞数目、肌细胞直径、肌细胞截面积以及胶原纤维截面积占比。对肌细胞中α-肌动蛋白(α-actin-1)、α-辅肌动蛋白(α-actinin-2)进行免疫组织化学染色共同反映肌细胞中收缩相关蛋白代谢变化。结果:①大鼠面肌形态学变化:神经断伤侧面肌肌细胞数目、肌细胞直径、肌细胞横截面积均小于对照侧(P<0.05)。神经断伤侧面肌中胶原纤维截面积占比大于对照侧(P<0.05)。②大鼠面肌收缩相关蛋白变化:面神经断伤侧面肌的α-肌动蛋白定量分析平均光密度小于对照侧,(P<0.05);面神经断伤侧面肌的α-辅肌动蛋白定量分析平均光密度与对照侧无明显差异,(P>0.05)。结论:大鼠面神经断伤2月后面肌萎缩明显,胶原纤维大量增生;肌细胞收缩功能减弱,肌细胞中蛋白质变化趋势不同,α-肌动蛋白含量下降,α-辅肌动蛋白含量未明显变化。 |

Objective: To observe the changes of facial muscle morphology and myocyte contraction-related proteins in rats after loss of innervation. Methods: In 23 healthy adult SD rats, the trunk of the facial nerve exiting the mammary foramen was cut on the left side (wounded side), and the facial nerve was exposed on the right side (control side). 2 months later, both nasolabial raphe muscles were taken and Masson staining was performed to observe the morphological changes of myofibers on both sides, including the number of myocytes, myocyte diameter, myocyte cross-sectional area and the percentage of collagen fiber cross-sectional area. Immunohistochemical staining of α-actin-1 and α-actinin-2 in myocytes was performed to reflect the metabolic changes of contraction-related proteins in myocytes. Results: (1) Morphological changes of rat facial muscles: the number of myocytes, myocyte diameter and myocyte cross-sectional area of the nerve-injured lateral muscles were smaller than those of the control side (P < 0.05). The cross-sectional area of collagen fibers in the nerve-injured facial muscle was larger than that of the control side (P < 0.05). The mean optical density of α-actin in the facial nerve injured lateral muscle was smaller than that in the control side (P < 0.05); the mean optical density of α-actinin in the facial nerve injured lateral muscle was not significantly different from that in the control side (P > 0.05). Conclusion: The muscle atrophy and collagen fiber proliferation were obvious after 2 months of facial nerve injury in rats; the contractile function of myocytes was weakened, and the trend of protein changes in myocytes was different, the content of α-actin decreased, and the content of α-actinin did not change significantly. |
