目的:研究富硒治疗对急性一氧化碳(CO)中毒性脑病模型大鼠的作用及其作用机制。方法:60只SD
大鼠随机分为正常对照组(BC组)、急性CO中毒组(CO组)、低硒治疗组(L-Se组)、中硒治疗组(M-Se组)、
高硒治疗组(H-Se组)各12只。腹腔注射CO制备急性CO中毒模型。造模前30 d及造模后每天分别给予各
组纯水或不同剂量富硒药物灌胃治疗。造模后第7天、第14天分别取脑及外周脏器,HE染色观察组织变
化;对脑皮质行小白蛋白(PV)免疫组化染色,Western Blotting检测皮质PV和Caspase-3的蛋白表达水平。
结果:硒对急性CO中毒大鼠模型的肝脏、肾脏无明显的毒性损伤。H-Se组在造模后第7天、第14天的皮质
PV阳性细胞数均高于CO组(P<0.05);L-Se组、M-Se组在造模后第7天、第14天的皮质PV阳性细胞数与
CO组无明显差异(P>0.05);H-Se组的PV阳性细胞数与BC组无明显差异(P>0.05)。H-Se组在造模后第7
天、第14天的PV表达量高于CO组(P< 0.05),Caspase-3蛋白表达低于CO组(P<0.05),与BC组无明显差异
(P>0.05);L-Se组、M-Se组在造模后第7天、第14天的PV和Caspase-3蛋白表达量与CO组无明显差异(P>
0.05)。结论:高剂量的硒治疗可明显上调PV,减少Caspase-3的活化。 |

To investigate the potential mechanism and protective effect against brain tissue damage
in rats by selenium-rich treatment after acute carbon monoxide(CO) poisoning. Methods: A total of 60 SD rats
were randomly assigned to groups blank control(BC), CO poisoning(CO), low dose selenium treatment(L-Se),
medium dose selenium treatment group (M-Se), and high dose selenium treatment group (H-Se), with 12 rats per
group. Rats were intraperitoneally injected with CO to establish animal models of acute CO poisoning while
those in the control group were injected with an equal volume of air. 30 days before and every day after generating model, rats were treated with H2O or various doses of selenium-rich drug. 7 and 14 days after generating model, cerebral cortical tissue and peripheral tissues were collected, HE staining was performed on peripheral tissue
organs to observe structural changes. Immunohistochemistry and Western blot were performed on cortical tissue
to assess levels of parvalbumin (PV) and Caspase-3 protein expression. Results: CO poisoned rat peripheral tissues sustained no significant injury from selenium treatment. The average PV positive cells in the 7-day and
14-day poisoning groups were significantly reduced compared to the H-Se group (P<0.05); reduction in the 7-day
and 14-day poisoning groups was slightly significant compared to those in the L-Se group and M-Se group (P>
0.05). There was no significant difference in PV positive cells between the BC group and H-Se group. PV level
in the 7-day and 14-day H-Se treatment groups was significantly higher than that in the CO group(P<0.05) and
Caspase-3 level was lower than the CO group (P<0.05). There was no significant difference in PV and Caspase-3
level between the 7-day and 14-day H-Se treatment groups and the BC group (P>0.05). There was no significant
difference in PV and Caspase-3 level between groups L-Se, M-se and CO group at 7-day and 14-day (P>0.05).
Conclusion: High doses of selenium treatment can markedly increase the level of PV and reduce the activation
of Caspase-3 |
